摘  要:目的利用25对AFLP引物组合对DDK品系小鼠与实验室常用的3个小鼠品系进行研究,分析DDK品系小鼠的遗传种质性,为解决其独特繁殖现象与遗传背景的关系做好理论准备。方法根据AFLP分析结果,统计每个引物组合在各个品系中的多态性条带数和特异性条带数,计算DDK小鼠与其他实验室常用小鼠(C57BL/6J、BALB/c、PWK)之间的遗传相似系数和遗传距离。结果 25对AFLP引物组合共检测到1 224个条带,多态性条带有279条,占总条带的22.79%。结论 DDK品系小鼠与BALB/c小鼠品系遗传关系最近,与C57BL/6J品系关系较近;DDK品系小鼠与PWK品系小鼠遗传关系最远;本研究为进一步探讨DDK小鼠的独特繁殖现象提供了理论依据;同时也阐明了AFLP技术在物种鉴定、遗传背景分析及预测亲本杂交效果等方面具有明显的优越性。Objective To analyze the genetic quality of DDK mice,we selected 3 strains commonly used in laboratory besides DDK mice for our study which may address the unique propagation phenomena of DDK mice and the relationship among the genetic backgrounds of 4 strains.25 pairs of AFLP primer combinations were selected to do the experiment.Methods According to AFLP results,we statisticed the number of polymorphic bands and the specific bands in each primer combination in all strains for calculating the genetic similarity coefficient and genetic distance among 4 strains of mice(DDK,C57BL/6J,BALB/c,and PWK).Results A total of 1,224 bands were detected by 25 pairs of AFLP primer combinations,which included 279 polymorphic bands,accounting for 22.79% of the total bands.Conclusions DDK strains of mice have a closest genetic relationship with BALB/c mouse strain,followed by the C57BL/6J strain.The genetic relationship between DDK mice and PWK strain mice is the farthest.This study provides a theoretical basis for further studying the unique breeding phenomenon of DDK mice;meanwhile,it clarifies that AFLP technology has the obvious advantages in species identification,genetic background analysis and prediction of effects such as hybrid parents.
