摘 要:目的对榆耳粗多糖(GI)进行分离纯化,并对所得均一多糖进行结构鉴定及抗肿瘤活性的研究。方法 GI经Q-Sepharose F.F离子交换色谱,Sepharose CL-6B凝胶色谱分离得到均一多糖,采用凝胶色谱、薄层色谱、红外光谱、高碘酸氧化等方法测定相对分子质量(Mr)、单糖组成、糖苷键连接方式。采用MTT法对粗多糖和均一多糖的体外抗肿瘤活性进行测定。结果分离得到均一多糖GI1-3和GI1-4b,其Mr分别约为1.55×105和3.03×104。经薄层色谱法、红外光谱法、高碘酸氧化法确定GI1-3由半乳糖、葡萄糖、甘露糖、木糖组成,以β-(1,4)、β-(1,6)、β-(1,3)糖苷键连接。GI1-4b由葡萄糖、甘露糖和木糖组成,以β-(1,4)、β-(1,6)糖苷键连接。体外抗肿瘤实验表明,GI1-3的抑瘤活性较GI1-4b和GI高,GI的抑瘤活性在3种多糖组分中最低。结论 GI、GI1-3和GI1-4b对体外生长的肿瘤细胞具有不同的抑制作用,可能与其Mr及分子结构有关。Purpose Gleoestereum incarnatum crude polysaccharide was selected to be purified,to conduct structural identifiction and to carry on research on antitumor activity.Methods The homogeneous polysaccharides were obtained by the way of Q-Sepharose F.F and Sepharose CL-6B,and they were identificated by thin layer chromatography,infrared spectrometry and periodate oxidation method.The antitumor activity was determined by MTT methods.Results Molecular weights of GI1-3 and GI1-4b were about 1.55×105 Da and 3.03×104 Da.GI1-3 was made of galactose,glucose,mannose and xylose,linking to β-(1,4),β-(1,6),β-(1,3) glycosidic bond.GI1-4b was made of glucose,mannose and xylose linking to β-(1,4),β-(1,6) glycosidic bond.The result of MTT showed that GI1-3 was the best component having antitumor activity,and GI was the worst one of three polysaccharides which we tested.Conclusion GI,GI1-3 and GI1-4b have different inhibiting effect on tumor cell in vitro,and this may be related to the difference of their molecular weight and molecular structure.
