摘  要:从兰州石化污水处理厂的活性污泥中分离二株能以苯酚为惟一碳源和能源生长的菌株,分别命名为lsd03,lsd05.通过逐级驯化,可在1000 mg/L苯酚的培养基中良好生长.利用聚合酶链式反应(PCR)方法和琼脂糖凝胶电泳技术检测到菌株中均含苯酚羟化酶基因片段的特异性条带,从基因水平上证实了lsd03,lsd05均具有降解苯酚的遗传基础.对这两株菌进行形态特征以及16S rDNA序列比对分析,初步确认了其在微生物系统发育学上的地位,均属于假单胞菌属(Pseudomonas).通过浊度测定法对菌株lsd03,lsd05在苯酚选择性培养基中的生长特性进行了研究,结果表明:其达到对数期的时间均为24 h,均能在1 000 mg/L的苯酚质量浓度下良好生长,lsd03的最佳培养温度为30~35℃,最佳培养pH环境为中性偏酸性;lsd05的最佳培养温度范围较广,在20~35℃均能良好生长,最佳培养pH环境为中性偏碱性.Two strains, named Petrochemical Sewage Disposal lsd03, lsd05 respectively, were isoIated Plant. They had the ability to utilize from the activated sludge of Lanzhou phenol as the sole carbon and energy sources. With the step-by-step domesticating method, they could grow well in the culture medium by using 1 000 mg/L phenol as sole carbon source, lsd03, lsd05 contained the specific bands of phenol hydroxylase gene fragment, which were detected by PCR and agarose gel electrophoresis, thus confirming the genetic basis of a degradation of phenol in the genetic level. They were considered to belong to Pseudomonas by morphological characterization and alignment analysis of the 16S rDNA sequence. The growth characteristics of lsd03 and lsd05 were studied by turbidity assay and the results showed that the time for both to reach the logarithmic period was 24 h and both were able to grow well at a concentration of 1000 mg/L phenol. The best culture temperature for lsd03 is 30-35 ℃ and the best culture pH environment is neutral and acidic: the best culture temperature for lsd05 is 20-35 ℃ and the best culture pH environment is neutral and alkaline.
