摘  要:目的筛选大孔树脂吸附法分离橄榄总黄酮的最佳条件。方法采用动态吸附-解吸实验筛选最佳大孔树脂,并以总黄酮含量为考核指标,用紫外分光光度法测定总黄酮含量,来确定最佳工艺条件。结果最佳工艺条件为:采用使用D101型大孔树脂,配制pH4.5吸附液以2倍柱体积/h上柱,至泄漏。用3倍柱体积纯水以2倍柱体积/h洗柱后,再用3倍柱体积60%乙醇洗脱,流速为1倍柱体积/h,将洗脱液减压浓缩,干燥得橄榄总黄酮。结论该法简单可行,分离效果好,可为工业生产提供参考数据。Objective To study the optimal separation conditions for the total flavonoids from Chinese olive with appropriate macroporous resin.Methods Dynamic adsorption-desorption methods were adopted to screen the optimal macroporous resin.The separation conditions were evaluated by measuring the concentration of the total flavonoids with UV-Vis spectrophotometer.Results The optimum conditions were as following:the type of macroporous resin was D101,the sample fluid was at pH 4.5 with the velocity of twofold of bed volume(BV)per hour,the water was threefold of bed volume with velocity of 2 BV/h,and 60% ethanol was threefold of bed volume with velocity of 2 BV/h.Conclusion This procedure is good and simple,and could be used to separate and purify the total flavonoids from Chinese olive.
