地塞米松对大鼠支气管哮喘气道重塑及PDGF-BB表达的影响

摘  要:目的观察大鼠支气管哮喘气道重塑模型中血小板源生长因子-BB(Platelet-derived growth factor-BB PDGF-BB)的表达变化,探讨了PDGF-BB与气道重塑的关系及早晚期使用地塞米松对支气管哮喘气道重塑和PDGF-BB表达变化的影响。方法将40只Wistar大鼠随机分为对照组(C组)、模型组(N组)、早期干预组(E组)和晚期干预组(L组),每组10只。N组、E组、L组每只大鼠于实验1、8天腹腔注射10%卵白蛋白抗原混悬液致敏,第15天始超声雾化1%卵白蛋白吸入8周20min/d激发哮喘,制备哮喘气道重塑模型。其中,E组、L组每只大鼠分别于雾化的第1、15天始,在每次雾化前1h腹腔注射地塞米松0.5mg/kg。C组以等量的生理盐水代替。肺组织切片HE染色,计算机图像分析气道形态学参数,衡量气道重塑的程度;免疫组化法测定PDGF-BB的表达。数据用SPSS16.0统计学软件进行处理。结果 E组、L组、N组均有不同程度的气道重塑,E组气道重塑程度明显较C组高且低于N组、L组(P<0.05)。PDGF-BB的表达E组较C组高且低于N组、L组(P<0.05)。PDGF-BB的含量与气道重塑程度相关(P<0.001)。结论 PDGF-BB与支气管哮喘气道重塑程度相关,在气道重塑中起重要作用。早期使用地塞米松可延缓但不能逆转气道重塑。Objective To observe the expression of PDGF-BB in airway remodeling model of asthmatic rats,explore not only the relationship between PDGF-BB and airway remodeling but also the effects of airway remodeling of asthma and expressing of PDGF-BB with the dexamethasone in the early and late phase. Methods Forty wistar rats were randomly divided into 4 groups :control group(C group);asthma group(N group);earty-interventional group(E group);late-inerventional group(L group). Each group contained ten rats.Each rats which were N group. E group and L group used intraperitoneally injection with 10% ovalbumin antigen suspension to sensitize on the first and eighth days in the study and began from the fifteenth day ultrasonic atomizing inhalation with 1% ovalbumin eight weeks and each day 20min to stimulate asthma,to establish the asthma model.Began from the first day and fifteeth day of ultrasonic atomizing inhalation,each rat in the E group. N group and L group intraperitoneally injected into dexamethasone(0.5mg/kg) one hour before each ultrosonic atomizing inhalation.C group was replaced by sodium chloride.Lung tissue were sliced and hematoxylin and eosin stained.The airway morphological parameters was measured by computer image analysis system to estimate the degree of airway remodeling.Expression of PDGF-BB was measured by immunohistochemical method.The data was analysed by spss 16.0. Results E group,L group and N group had different levels of airway remodeling. The degree of airway remodeling in the E group was significantly thicken than the C group and thinner than the N group and L group(P0.05).Expression of PDGF-BB in the E group was higher than the C group and lower than N group and L group(P0.05).Expression of PDGF-BB correlated with the degree of airway remodeling(P0.001).Conclusion Expression of PDGF-BB was related with the degree of airway remodeling of asthma and may play an important role in airway remodeling.Using dexamethasone in early phase can delay but not reverse airway
