摘  要:目的:建立高效液相色谱法测定大鼠血清中马钱苷的浓度,研究大鼠灌胃给药后体内药代动力学特点。方法:采用KromasilC18色谱柱(4.6mm×250mm,5μm),流动相:0.05%磷酸水溶液(A)-乙腈(B),流速1.0mL/min,检测波长:236nm。结果:马钱苷回归方程:Y=1.0921X+27.064,r=0.9991,线性范围为26~420ng;平均回收率为94.20%,RSD=4.01%;样品24h内稳定,RSD=0.55%。大鼠灌胃后,药动学行为符合一室模型,主要药动学参数Ka、Ke、t1/2Ka、t1/2Ke、Tpeak、Cmax、AUC分别为0.0085/min、0.025/min、27.95min、81.23min、65.59min,6970.40ng/mL,1429620.62ng/(mL·min)。结论:首次建立了浓缩六味地黄丸中马钱苷血药浓度的HPLC测定方法,阐明了六味地黄丸中马钱苷的药代动力学参数,方法专属性强,结果可靠。AIM:To establish a revered-phase HPLC method for studying pharmacokinetics of loganin in serum of rat after perfusion.METHODS:The Kromasil C18(4.6 mm × 250 mm,5 μm)column was used as the analytical column and the mobile phase consisted of 0.05% phosphoric acid solution and acetonitrile,with the flow rate at 1.0 mL/min).The UV detection was set at 236 nm.RESULTS:The calibration curves of loganin was linear in the range of 26 ~ 420 ng(r = 0.999 1).The recovery and relative standard deviation of loganin was 94.20% and 4.01%,respectively.The loganin-containing serum was not changed in 24 hours(relative standard 0.55%).After a perfusion of Liuweidihuang Concentrated Pill,the mean plasma concentrations-time relationship was found to fit a one-compartment open model,the main pharmacokinetic parameters of loganin were as follows:Ka,Ke,t1/2Ka,t1/2Ke,Tpeak,Cmax,AUC were 0.008 5 /min,0.025 /min,27.95 min,81.23 min,65.59 min,6 970.40 ng/mL,1 429 620.62 ng/(mL·min).CONCLUSION:This is for the first time to establish the revered-phase HPLC method to determine concentrations of loganin in serum of Liuweidihuang Concentrated Pill and to obtain the pharmacokinetic parameters and characteristics.The method is reliable,the result is accurate.
