原发性高血压患者颈总动脉超声等临床变量与髓过氧化物酶基因变异的关系

摘　　要：目的:探讨原发性高血压(EH)患者颈总动脉超声等临床变量与髓过氧化物酶(MPO)基因变异的关系。方法:应用聚和酶链反应(PCR)检测EH组107例及对照组97例患者MPO基因型,比较不同基因型之间的分布频率。同时使用B型彩色多普勒超声等检测2组患者的颈总动脉超声及其他临床变量,并进行对照分析。结果:正常人群GG、GA、AA基因型频率分别为56.7%、40.2%和3.1%,EH病例组分别为70.1%、29.0%和0.9%,EH组MPO基因AA+GA基因型频率低于对照组,二者比较有差异性(P=0.047)。EH患者的颈总动脉内径、内膜厚度等临床变量的变化与MPO不同基因型相关。结论:本研究人群MPO基因多态性与EH遗传易感性相关。EH患者的颈总动脉内径、内膜厚度及各临床变量的变化与MPO基因463G/A的变异有关。Objective：Relationship between MPO gene polymorphism and carotid artery uitrasound as well as clinical variables in essential hypertension.Method：MPO genotypes in 107cases of essential hypertension and 97 persons of healthy control were detected using PCR-restriction fragment length polymorphism assay（PCR-RFLP） in a case-control molecular epidemiology study.MPO gene polymorphism with carotid artery uiltrasound as well as clinical variables were compared between two groups.Result：In healthy control group,the frequencies of persons carrying G/G,G/A,and A/A genotypes were 56.7%,40.2%and 3.1%,respectively.In essential hypertension group,the frequencies of the persons carrying above three genotypes were 70.1%、29.0%and 0.9%,respectively.The frequency of MPO genotypes（AA＋GA）in hypertensive was significant difference than in control.The MPO gene polymorphism was associated with carotid artery uiltrasound and the clinical variables in EH.Conclusion：MPO gene polymorphism was associated with susceptibility in EH.Relationship between MPO gene polymorphism and carotid artery uitrasound as well as clinical variables in EH.
