摘  要:目的:观察氧化低密度脂蛋白(ox-LDL)对血管平滑肌细胞(VSMC)血管紧张素Ⅱ受体-1(AT1R)表达的影响及凝集素样氧化低密度脂蛋白受体-1(LOX-1)在其中的介导作用。方法:人脐带动脉平滑肌细胞体外原代培养。ox-LDL干预,RT-PCR测定VSMCAT1R mRNA和LOX-1mRNA表达。并观察在LOX-1的抑制剂多聚肌苷酸作用后,VSMCAT1R mRNA表达的变化。结果:①20mg/Lox-LDL作用6h、12h及24h后,VSMCAT1R mRNA表达升高,与空白对照组比较,均P<0.01,作用6h出现峰值。②1、10、100mg/L浓度ox-LDL作用后,VSMCLOX-1mRNA表达均升高,与空白对照组比较,P<0.05~P<0.01;且随着ox-LDL浓度的升高有升高趋势,各浓度之间比较,P<0.01。③多聚肌苷酸抑制LOX-1后,AT1Rm-RNA表达明显下降,与空白对照组、ox-LDL单独作用组比较,均P<0.01。结论:ox-LDL可持续性促进VSMCAT1R表达,LOX-1在此过程中起重要介导作用。Obstract:The purpose of this study is was to observe,in the inflammation course,how ox-LDL impact AT1R m-RNA expression of VSMC and how LOX-1 act as.Method:Smooth muscle cells from human umbilical artery was were cultured in vitro.Grouping according to different ox-LDL concentrations and reaction times.Then,detection the quantity of VSMC AT1R m-RNA and LOX-1 m-RNA with RT-PCR.Further more,observe the influence of polyinosinic acid,one inhibitor of LOX-1,on the production of VSMC AT1R m-RNA.Result:①The quantity of VSMC AT1R m-RNA increased highly after co-cultured with ox-LDL,P0.01.The peak value was found at about 6th hours.The expression of AT1R m-RNA was increased as ox-LDL concentration ascending.②The quantity of VSMC LOX-1 m-RNA increased significantly with dependence on ox-LDL concentration.③AT1R m-RNA was reduced significantly when LOX-1 was inhibited by polyinosinic acid.Conclusion:ox-LDL can induce increased expression of VSMC AT1R m-RNA.LOX-1 plays a vital rule in this process.
