家蚕病原白僵菌原生质体的再生回复研究

摘  要:研究了家蚕病原白僵菌(Beauveriabasiana)原生质体的性状、再生回复形式与条件。家蚕病原白僵菌原生质体的无核率为265%,有核率为735%,其中单核率为535%。原生质体再生回复的形式可观察到3种,在位相差显微镜下不能判别原生质体有无再生回复能力。综合考虑原生质体再生频率和菌落生长发育速度,家蚕病原白僵菌原生质体再生回复用培养基的渗透压稳定剂以07mol/L葡萄糖较为适当。Regeneration and reversion of protoplasts from pathogenic fungus, Beauveria bassiana of silkworm, Bombyx mori L.were studied.Among the protoplasts,26.5% were without a nucleus,53.5% contained a single nucleus,and the other protoplasts contained two or more nuclei.Three morphologically distinct regeneration and reversion types of the potoplasts were observed,and the regenerating protoplasts could not be distinguished from nonregenerating protoplasts under a phase contrast microscope.Through the comprehensive survey of the colony formation and growth of the protoplasts,it is suggested that glucose of 0.7 mol/L as osmotic stabilizers for regeneration and reversion of the protoplasts is more suitable.
