摘  要:目的通过大鼠脑皮质神经元的体外培养,观察遭受机械性离心损伤时,米诺环素对神经元的保护作用。方法通过变速离心培养板,造成神经元损伤的作用。使用倒置相差显微镜观察损伤后神经元的形态改变,并测量损伤前后不同时间段损伤组与损伤加米诺环素组乳酸脱氢酶(LDH)含量变化。结果光镜见轴索肿胀、轴索和神经元崩解。损伤组与对照组神经细胞损伤后LDH值比较差异有显著性(P<0.01);损伤后30 min损伤加米诺环素各组LDH的水平与损伤组比较均明显上升(P<0.05;P<0.01);损伤后24 h、48 h损伤加米诺环素中、低浓度组LDH的水平与损伤组比较均明显下降(P<0.05;P<0.01),高浓度组则无明显影响。结论米诺环素对机械性损伤大鼠脑皮质神经元有双重作用,高、中、低浓度不能减轻神经元膜的原发性损伤,但中、低浓度能减轻神经元膜的继发性损伤。Objective To observe the role of minocycline in centrifugation-induced cortical neuron in vitro.Methods By means of centrifugating culture plate with vital neuron in vitro,the shape of axons was observed with microscope.Meanwhile lactate dehydrogenase(LDH) was measured in culture medium at different stages.Results Axonal swelling,disruption of axon and neuron were found under microscope.In injury group,LDH showed difference with that in control(P0.01).In each minocycline group 30min after injury,LDH showed significant difference with that in injury(P0.05;P0.01).In lower concentrations minocycline group 24h,48h after injury,LDH showed significant difference with that in injury(P0.05;P0.01).There was no significant effect in high concentration minocycline group.Conclusion Minocycline has double effects on centrifugation-induced rat cortical neuron.It can not prevent neuron from primary injury at different concentrations,but it can prevent neuron from secondary injury at lower concentrations.
