摘  要:目的探讨泥鳅分泌多糖对蛋白质非酶糖基化反应的干预作用。方法通过NBT还原实验、二羰基化合物测定实验、非酶糖基化终产物(advanced glycation end products,AGEs)的荧光性测定实验,以氨基胍为阳性对照,从糖基化反应的三个不同阶段分别测定泥鳅分泌多糖对糖基化的干预作用。用十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate-polyacrylamide gel electrophoresis,SDS-PAGE)检验泥鳅分泌多糖对糖基化反应的干预结果。结果泥鳅分泌多糖可以从糖基化反应的3个阶段来抑制AGEs的生成,表现出很强的抑制作用,尤其对二羰基化合物的生成抑制作用显著,在浓度为200μg/mL时,抑制率为57.4%,与阳性对照氨基胍比较无显著性差异(P>0.05)。电泳结果表明在分子量90 kD处出现明显的糖基化蛋白条带,泥鳅分泌多糖对糖基化蛋白生成的抑制作用与氨基胍比较无显著性差异(P>0.05)。结论泥鳅分泌多糖对蛋白质糖基化有显著的抑制作用。Objective To study the effect of polysaccharide from secretion of Misgurnus anguillicaudatus on antiglycation.Methods Through NBT reductive assay,measurement of dicarbonyl compounds and determination of the fluorescence of advanced glycation end products(AGEs),the effect of polysaccharide from secretion of Misgurnus anguillicaudatus on the three periods of non-enzymatic glycation reaction were investigated,with aminoguanidine as positive control.SDS-PAGE was performed to verify the effect of polysaccharide on glycation.Results The polysaccharide showed strong inhibitory effect during the three periods of the formation of AGEs,especially on the formation of dicabonyl compounds.At the concentration of 200 μg/mL,the inhibitory rate of the formation of dicabonyl compounds was 57.4%,with no significant difference compared with aminoguanidine(P0.05).There was an obvious glycated protein band(Mw=90kDa) in SDS-PAGE result,the inhibitory effect of the polysaccharide on AGEs was similar with aminoguanidine(P0.05).Conclusion Polysaccharide from secretion of Misgurnus anguillicaudatus has obvious inhibitory effect on the development of non-enzymatic glycation reaction.
