摘 要:目的探讨热休克预适应产生的热休克蛋白70(HSP70)在心脏移植供心中的表达规律和对心脏移植供心的影响。方法建立大鼠异位心脏移植模型并随机分为6组:对照组,经热处理的R0 h、R24 h、R48 h、R96 h、R192 h实验组。分别于移植后1周取出供心,测定其中HSP70含量、心肌组织生化指标变化并观察心肌组织超微结构的变化。结果 R24 h、R48 h组HSP70含量较其他4组明显升高,心肌组织生化指标也优于其他4组(P〈0.01),供体心肌超微结构的损伤也较其他4组明显减轻。结论热休克预适应可诱导大鼠心肌HSP70含量逐渐增加,并在24~48 h达高峰,一定量的HSP70表达对心脏移植的供心具有保护作用。Objective To research the expression of the heat shock protein 70(HSP70)which was generated by heat shock preconditioning and its influence to the donor's heart in the heart transplantation.Methods Rat model of heterotopic heart transplantation was developed and six groups were divided as following: control group,R0 h group,R24 h group,R48 h group,R96 h group,R192 h group.Hearts were harvested at post-operative one week and the content of HSP70,myocardial biochemical indicator,myocardial microstructural changes were detected.Results The peak of HSP70 was in R24 h group and R48 h group,and the myocardial biochemical indicators of them also better than those of the other 4 groups(P0.01).The two groups' myocardial microstructural injury also eased than the other four groups.Conclusion Heat shock preconditioning can induce the content of HSP70 increasing gradually in the rat myocardium and reach the peak at 24 to 48 h,a certain amount of HSP70 expression has protective effect on the donor's heart in the heart transplantation.
