摘  要:将旋毛虫TsP53重组蛋白以20μg/只的剂量免疫小鼠3次后攻击感染旋毛虫肌幼虫200条/只。检查小鼠肠道旋毛虫7日龄成虫数、雌虫体外产新生幼虫数和感染35d后的肌幼虫数,并计算减虫率,同时用间接ELISA检测血清中抗TsP53重组蛋白的抗体应答。结果显示,TsP53重组蛋白免疫小鼠旋毛虫7日龄成虫、雌虫产新生幼虫的减虫率分别为14.34%和16.93%,与感染对照组和佐剂对照组相比差异不显著(P>0.05);感染35d后肌幼虫减虫率为38.68%,与感染对照组和佐剂对照组相比差异显著(P<0.05)。TsP53重组蛋白免疫小鼠在首次免疫后第7天出现抗体应答,可检测到抗TsP53重组蛋白的抗体,第3次免疫后第7天达到峰值,攻击感染后略有下降,但直到试验结束时仍然维持在较高水平,TsP53重组蛋白可诱导小鼠产生部分抗旋毛虫的免疫保护性。Forty mice in immunization group were immunized with 20μg TsP53recombinant protein 3 times and 30mice in adjuvant group and 30mice in infection control group were challenged with 200infective larvae of Trichinella spiralis per mouse on day 7post-3rd-immunization.The reduction rates of 7-day-old adult worms,production of newborn larvae of female adult in vitro and muscle larvae on day 35post-in- fection were counted and the anti-TsP53IgG level in the sera of immunized animals was detected by indi- rect ELISA.The reduction rate of 7-day-old adult worms,newborn larvae and muscle larvae were 14.34%, 16.93%and 38.68%,respectively.TsP53-specific antibody response in the immunized mice was detectable on day 7post-1st-immunization and reached the peak on day 7post-3rd-immunization.The anti-TsP53 IgG level in the immunized mice was significantly higher than that in the control groups.The result showed that the TsP53 recombinant antigen could induce partial immune protection in mice against T.spiralis.
