摘  要:目的探讨罗格列酮对抵抗素基因转染的HepG2细胞转录因子PPARγ mRNA表达和活性氧的影响。方法用微量化的葡萄糖氧化酶法对抵抗素基因转染诱导的胰岛素抵抗细胞进行鉴定,加用罗格列酮干预,并和正常HepG2细胞进行对照,用流式细胞仪检测细胞活性氧和RT-PCR法检测PPARγ mRNA表达。结果抵抗素基因转染诱导了肝性胰岛素抵抗的细胞模型,罗格列酮对抵抗素转染的HepG2细胞PPARγ mRNA表达有显著的抑制作用,对活性氧含量改变影响不明显。结论糖尿患者长期服用罗格列酮可能对肝脏PPARγ mRNA的表达有抑制作用。【Objective】 To explore the effects of Rosiglitasone on the PPAR γmRNA and reactive oxidative species content in resistin gene transfected HepG2 cells.【Methods】To identify the hepatocyte insulin-resistant model in resistin gene transfected HepG2 cells with micro GOD-POD,interference with Rosiglitasone and compared with control,to detect the content of reactive oxidative species with flow cytometry and expression of PPARγ mRNA with RT-PCR.【Results】 Resistin gene transfection on HepG2 cells induced insulin resistance hepatocyte model,Rosiglitasone application in resistin gene transfected cells significantly down-regulated the expression of PPARγ mRNA,but the changes of reactive oxidative species content was not obvious.【Conclusion】Long-term administration of Rosiglitasone in patients with Diabetes Mellitus may suppress the hepatic PPARγ mRNA.
