摘 要:目的观察局灶性脑缺血再灌注大鼠脑皮质区组织基质金属蛋白酶-9(MMP-9)的表达及川芎嗪缓释剂穴位埋药线和川芎嗪穴位注射干预对其的影响。方法将SD大鼠随机分为正常组、假手术组、模型组(1d组、3d组、5d组)、穴位注射组(1d组、3d组、5d组)、穴位埋药线组(1d组、3d组、5d组)、尾静脉注射组(1d组、3d组、5d组)。采用改良线栓法制备局灶性脑缺血(MCAO)再灌模型。穴位埋药线组在大椎、百会穴埋入胶原川芎嗪缓释剂药线;穴位注射组在大椎、百会穴注射川芎嗪注射液;运用蛋白免疫印迹法检测缺血侧脑皮质区组织MMP-9蛋白的表达和含量。结果穴位埋药线组与穴位注射组能明显下调缺血侧脑皮质中MMP-9的表达和含量,与模型组比较差异有统计学意义,其中穴位埋药线组的下调作用最明显,与穴位注射组相比较差异有统计学意义。结论穴位埋药线可下调脑缺血再灌注大鼠缺血侧皮质组织MMP-9的表达,进而可能减少脑微血管基底膜的降解,保护微血管的完整性,减少脑缺血再灌注损伤。Objective: To observe the MMP-9 expression and concerntration in cortex area of rats with cerebral ischemia.And to discuss the effect of embedding medicine in acupoint and point injection with tetramethylpyrazine in this disease Methods: 72 SD rats were randomly divided into the normal group,the sham operation group,the model groups(the 1d group,the 3d gruop and the 5d group),the acupoint injection group(the 1d group,the 3d gruop and the 5d group),the embedding medicine in acupoints group(the 1d group,the 3d gruop and the 5d group)and the tail vein injection group(the 1d group,the 3d gruop and the 5d group).The model of cerebral artery occlusion(MCAO) and reperfusion was developed by occluding rat’s right middle cerebral artery following suture occlusion method.After establishing the model successfully,the embedding medicine in acupoints group was implanted into Baihui(GV 20) and Dazhui(GV 14).The injecting medicine in acupoints group was injected into Baihui(GV 20) and Dazhui(GV 14).The content of MMP-9 in cortex tissues was evaluated with western blotting.Results: The content and the expression of MMP-9 in cortex tissues of model groups was significantly higher than that of other three groups.The content of MMP-9 in the cortex tissues of the groups of embedding medicine in acupoints,comparing with the groups of injecting medicine into rats’tail veins and of injecting medicine in acupoints,was cut down signifficantly.Comparing the groups of injecting medicine into rats’tail vein and the groups of injecting medicine in acupoints,the expression of MMP-9 of the frontier was less than the laster.Conclusion: The method of embedding medicine in acupoints could inhibit the expression of the MMP-9 in cortex tissues of rats with focal cerebral ischemia reperfusion injury to protect the brain and decrease the reperfusion injury.
