摘 要:分别对3种快速测定γ-氨基丁酸(γ-aminobutyric acid,简称GABA)含量的方法进行了研究:基于Berthelot反应的比色法、纸色谱萃取分光光度法、纸电泳分光光度法。首先探讨了3种方法各自的最佳测定条件,然后以马铃薯为对象,研究各种测定方法在测定提取液中GABA含量的效果,最后与氨基酸自动分析仪的测定结果作比较。结果表明:纸电泳分光光度法不仅操作简单、价格低廉,而且精确度高、重现性好,是一种适合于大批量进行马铃薯提取液中GABA含量快速测定的有效方法。Three simple methods of determining GABA were studied in this paper. They are the Berthelot spectrophotometry, paper chromatography and paper electrophoresis. The optimum conditions of each method were discussed, which were used to analyse the GABA content in potatoes. Then the results of three methods were respectively compared to that obtained by means of amino acids analyzer. It was showed that the paper electrophoresis was a simple and economical way with high sensitivity, accuracy and reproducibility, and suggested as an effective method for quickly detecting GABA in large numbers of potato samples.
