大鼠非梗死区心肌C3G蛋白的表达及苯肾上腺素对其的干预

摘　　要：目的:探讨非梗死区心肌C3G蛋白的表达及苯肾上腺素对其的干预。方法:选择雄性SD大鼠32只。按Litwin方法建立心肌梗死(简称心梗)及假手术模型。术后3天仍存活的雄性SD大鼠分为假手术组、心梗组、假手术苯肾上腺素组和心梗苯肾上腺素组(各组n=8)。假手术组及心梗组给予生理盐水1ml/(kg.d),假手术苯肾上腺素组及心梗苯肾上腺素组给予苯肾上腺素0.65mg/(kg.d),腹腔注射,至干预后8周。干预后应用免疫印迹法检测非梗死区心肌C3G蛋白的表达。结果:干预后12周,非梗死区心肌C3G蛋白表达积分光密度值(免疫印迹)分别为:假手术组(0.33±0.06),心梗组(0.81±0.05),假手术苯肾上腺素组(0.40±0.06),心梗苯肾上腺素组(0.62±0.07)。与假手术组比较,心梗组、心梗苯肾上腺素组非梗死区心肌C3G蛋白表达均增加,差异均有统计学意义(P<0.05);心梗苯肾上腺素组较心梗组非梗死区心肌C3G蛋白的表达显著降低,差异有统计学意义(P<0.05);而假手术苯肾上腺素组较假手术组非梗死区心肌C3G蛋白表达差异无统计学意义。结论:非梗死区心肌C3G蛋白的表达显著增加,而苯肾上腺素可显著调低其表达,提示C3G蛋白与梗死后心室重塑有关。Objective： To investigate C3G protein expression in non-infarcted myocardium with its phenylephrine （PE） regulation in rats. Methods： The myocardial infarction （MI） and sham-operation models were established using Litwin＇ s method in Sprague- Dawley male rats. 3 days after the operation,surviving rates were divided into 4 groups as Sham operation group,MI group,Sham ＋ PE group and MI ＋ PE group, n = 8 in each group. Animals in MI group and Sham operation group were treated with normal saline 1 mL/kg/day ; in Sham ＋ PE group and MI ＋ PE group were treated with PE 0. 65 mg/kg/day intraperitoneally for 8 weeks. All animals were fed for 12 weeks, and then, C3G protein expression in non-infarcted myoeardium was examined by Western blot. Results： 12 weeks after the treatment ,Western blot analysis presented that the integration optical density of C3G protein in Sham operation group, MI group, Sham ＋ PE group and MI ＋ PE group were （ 0. 33 ±0. 06,0. 81 ±0. 05,0. 40 ± 0. 06, and 0. 62 ± 0. 07） respectively. Compared with Sham operation group, C3G protein expression was increased in MI group （P 〈 0.05）, and compared with Sham ＋ PE group, C3G expression was higher in MI ＋ PE group （ P 〈 0. 05 ）. C3G protein expression was decreased in MI ＋ PE group than that in MI group（ P 〈0.05 ）. C3G expression was no difference between Sham ＋ PE group and Sham operation group. Conclusion： C3G protein expression was increased in non-infarcted myocardium in rats ,PE could down-regulate this expression. The relationship between C3G protein and left ventrieular remodeling had been confirmed.
