摘  要:目的探讨O139群、O1群霍乱弧菌分子遗传特征及其相互关系。方法用聚合酶链反应(PCR)、DNA序列分析及随机扩增多态性DNA(RAPD),检测了3株O139群、3株O1群ElTor生物型和2株古典生物型霍乱弧菌。结果3株O139群和5株O1群霍乱弧菌均扩增出566bp的DNA片段,DNA序列源于霍乱肠毒素(ctx)A2B基因。RAPD产生的两种DNA指纹图谱一致显示O139群与O1群ElTor生物型遗传特征极其相似,与古典生物型略有区别。结论O139群与O1群ElTor型存在密切的种系进化关系,前者可能由O1群ElTor生物型进化而来。Objective To study the inherent characteristics of Vibrio cholera O139 and O1 serogroup and the relationship between them.Methods Polymerase chain reaction (PCR), DNA sequences analysis and randomly amplified polymorphic DNA(RAPD) were used to test 3 strains of O139 serogroup, 3 strains of O1 El Tor biotype and 2 strains of classical biotype.Results The specific 566bp DNA segments were amplified of 3 strains of O139 serogroup and 5 strains of O1 serogroup, which were consistent with the genomic sequence of ctx A2 B. The two kinds of DNA fingerprints of RAPD amplified respectively by two primers showed that the inherent characteristics of O139 were almost same with the O1 El Tor biotype, while slight different with O1 classical biotype. Conclusion O139 serogroup and O1 El Tor biotype kept close inherent identity in evolution, and it is possible for O139 to evolute from O1 El Tor.
