摘 要:应用微卫星标记技术鉴别商品中常见禽源性成份。采集市售鸡、鸭、鹅的组织和内脏作为试验对象,提取基因组DNA。从GenBank中初选3种动物的微卫星标记,设计引物,用PCR-凝胶电泳检测方法筛选出每种动物种间特异的微卫星标记。试验结果表明:在鸡、鸭、鹅源性成份中,鉴定鸡源性成份可采用鸡的微卫星标记MCW 174,扩增的目的片段为261 bp或270 bp;鉴定鹅源性成份可采用鹅的微卫星标记EU833383,扩增的目的片段为479 bp或490 bp;鉴定鸭源性成份首先采用鸭的微卫星标记AMU70,扩增的目的片段为252 bp,264 bp或271bp,然后采用鹅的微卫星标记EU833383,无扩增条带出现。该方法成功的应用于鸡的处理样品和鸭的熟食类样品的检测。该方法具有准确快速、简便的优点,适用于鸡、鸭、鹅源性成份的鉴别。The aim of this study was to develop a PCR assay for the identification of chicken,duck and goose derived materials by microsatellite markers.Genomic DNAs were extracted from above three animal species meat and internal organs.The specific primers were designed based on the microsatellite DNA markers selected from GenBank.The results revealed that marker MCW174 could be applied for the identification of chicken derived materials from three species by PCR with a 261 bp or a 270 bp amplified DNA fragment.The microsatellite marker EU833383 could identify goose derived materials from three species with a 479 bp or a 490 bp PCR amplification product.Microsatellite marker AMU70 amplified a 252bp,a 264 bp or a 271 bp DNA fragment in the duck derived materials from three species.The method was applied successfully in the detection of processed samples of chicken and baked or salted samples of duck as well.This method is accurate,quick and convenient for the identification of chicken,duck and goose derived materials.
