摘 要:目的探讨不同维持温度对冷冻保存异体动脉内皮细胞代谢活性、超微结构和移植通畅率的影响。方法采用二步冷冻步骤,选择4种不同的维持温度(-20℃、-40℃、-60℃、-80℃)对异体颈动脉进行冷冻,在液氮中保存48天后施行移植。观察方法包括四唑盐还原试验、透射电镜、动脉造影及病理检查。结果经-60℃维持温度冷冻后再保存于液氮中的异体动脉,其代谢活性和超微结构得到较好保持,移植后排斥反应减弱,90天通畅率87.5%。Objective To investigated the effects of different holding temperature on the metabolic activity,ultrastructure and patency of cryopreserved allograft arteries Methods Allograft arteries were stored in liquid nitrogen 48 days by a two step freezing schedule,in which different holding temperature( 20℃ 40℃ 60℃ and 80℃)were employed in four groups,and then implanted into femoral artery defects The allograft arteries were observed after cryopreservation using tetrazolium test and transmission electron microscope and analysed postoperatively by angiography and pathologic examination Results The metabolic activity and ultrastructure of cryopreserved allograft arteries were well preserved at the holding temperaturd 60℃;There was a 87 5% patency rate at 90 days for such allografts with a decreased immunologic reaction Conclusions 60℃ is the optimal holding temperature for the cryopreservation of allograft artery
