摘 要:目的观察秦皮甲素对高脂饮食诱导的实验性大鼠脂肪肝的防治作用。方法 48只SD大鼠,随机分为正常对照组、模型组、辛伐他汀组(5 mg.kg-1.d-1),秦皮甲素低剂量组(10 mg.kg-1.d-1)、中剂量组(30 mg.kg-1.d-1)、高剂量组(90 mg.kg-1.d-1)。高脂饲料饲养大鼠诱导脂肪肝,同时灌胃上述各组药物,连续12 wk。测定血浆三酰甘油(TG)、总胆固醇(TC)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)浓度;血浆、肝脏匀浆丙二醛(MDA)浓度,血浆亚硝酸根(NO2-)浓度和肝脏脂质蓄积情况。结果与模型组相比较,秦皮甲素低、中、高3个剂量组均可显著降低血浆TG、LDL的浓度(P<0.05),而对TC、HDL则无明显影响(P>0.05);同时,均可显著降低血浆MDA浓度(P<0.05);秦皮甲素中、高2个剂量组显著降低肝脏MDA浓度(P<0.05),而对血浆NO2-无明显影响(P>0.05)。病理切片显示秦皮甲素显著降低脂质在肝脏的蓄积。结论秦皮甲素对实验性脂肪肝有一定的防治作用。AIM To explore the preventive and therapeutic effects of esculin on fatty liver induced by high-fat diet in rats.METHODS Forty-eight SD rats were randomized into six groups: the control group,the model group,the positive control group(simvastatin 5 mg.kg-1.d-1),the low(esculin 10 mg.kg-1.d-1),and the middle(esculin 30 mg.kg-1.d-1) and the high dose(esculin 90 mg.kg-1.d-1) group.Except the control group,SD rats were fed with high-fat diet for 12 weeks to induce fatty liver.The plasma concentration of triglyceride(TG),total cholesterol(TC),high-density lipoprotein(HDL),low-density lipoprotein(LDL) was determined.The plasma concentration and the liver tissue content of malonaldehyde(MDA) were monitored by TBA assay.Griess reagent was used to determine the plasma nitrite(NO2-) concentration.The liver tissue paraffin sections were stained by HE method and the lipid accumulation in the liver was observed.RESULTS Compared with the model group,esculin significantly decreased plasma TG and LDL concentrations(P 0.05) without affecting the TC and HDL(P 0.05).The plasma concentration of MDA was significantly decreased after esculin treatment in three doses of esculin treated groups(P 0.05).The liver tissue content of MDA was also dramatically decreased in middle and high dose groups of esculin administration(P 0.05).Esculin showed no effect on plasma NO2-level(P 0.05).The HE staining revealed that esculin could inhibit high-fat diet induced lipid accumulation in the liver.CONCLUSION Esculin has a certain therapeutic effects on experimental fatty liver in vivo.
