摘 要:目的:研究电刺激对PC12细胞内泛素-蛋白酶系统(UPS)的作用,以期了解细胞外电刺激作用于神经细胞的作用机制.方法:通过细胞实验,对PC12细胞株分0μA、500μA、2mA、8mA进行不同电流强度电刺激每天3h,连续4d;分0、10、130Hz进行不同电流频率电刺激每天3h,连续3d.之后采用噻唑蓝(MTT)检测存活率、酪氨酸羟化酶(TH-16)免疫组织化学检测细胞的新陈代谢,以及免疫印迹法检测细胞内泛素化蛋白量的改变.结果:随着电流的增强,电刺激对PC12细胞的生长有抑制趋势;高频电刺激对PC12细胞生长有抑制作用,对PC12细胞的酪氨酸羟化酶的合成代谢有抑制作用;高频电刺激使细胞内泛素化蛋白数量发生改变.结论:高频电刺激引发局部神经元兴奋性降低和细胞内UPS功能障碍,改变了原来信号转导通路,可能是临床上深部脑刺激治疗帕金森病的机制.Objective: To investigate the effect of electrical stimulation on the ubiquitin-proteasome system (UPS) of the neu-ron-like cells, and to find out the cell and molecular mechanism of the effect of extracellular electrical stimulation in the cells. Methods:PC12 cells were stimulated with different intensity (0 μA, 500μA, 2 mA, 8 mA) for 4 days, 3 hours per day; and with different frequency (0 Hz, 10 Hz, 130 Hz) for 3 days, 3 hours per day. Then, the cellular viability was examined by MTT assey, the cellular metabolism was examined by the immunohistochemistry of TH-16, and the cellular ubiquitinated protein was detected by Western blot. Results: Electrical stimulation tended to inhibit the growth of PC12 cell with increase of the current. High frequency stimulation inhibited the growth of PC12 cell, obstructed the anabolism of tyrosine hydroxy- lase of PC12 cell, and influenced the amount of the ubiquitinated proteins. Conclusion: High frequency stimulation induces the decrease of excitability of neurons and the dysfunction of UPS of cells, which changes the signal transduction pathway, and may play an important role of deep brain stimulation in Parkinson's disease.
