摘  要:目的探讨人参皂甙Rd对正加速度(+Gz)暴露后大鼠运动和学习记忆能力的影响。方法 64只雄性SD大鼠,体质量200~250g,随机均分为假手术组(Sham组)、正加速度组(+Gz组)、人参皂甙Rd处理组(Rd组)和溶剂对照组(Vehicle组)。Sham组大鼠静置于离心机上但不给予+Gz暴露;+Gz组大鼠给予+10Gz,5min暴露;Rd组大鼠在+10Gz,5min暴露前30min腹腔注射人参皂甙Rd30mg/kg;Vehicle组大鼠在+10Gz,5min暴露前30min腹腔内给予等容量溶剂(20%丙二醇)。+10Gz,5min暴露后不同时间点,通过旷场实验观察各组大鼠运动能力的变化;通过水迷宫实验观察各组大鼠学习记忆能力的变化。结果在旷场实验中,Rd组大鼠的站立次数较+Gz组明显增多(P<0.05)且在中央格的停留时间也明显减少(P<0.05)。而Vehicle组大鼠的站立次数、中央格的停留时间与+Gz组比较无显著性差异(P>0.05)。在水迷宫实验中,与+Gz组比较,Rd组的逃避潜伏期明显缩短(P<0.05);目标象限停留时间显著延长(P<0.05)。而Vehicle组大鼠的逃避潜伏期、目标象限停留时间与+Gz组比较无统计学差异(P>0.05)。结论人参皂甙Rd能改善+Gz导致的运动和学习记忆能力下降。Objective To investigate the effect of ginsenoside Rd on the impairment of locomotivity and ability of learning and memory induced by +Gz in rats.Methods A total of 64 male SD rats weighing 280~320 g were randomly divided into four groups. The rats in sham group were put on the animal centrifuge without exposure to +Gz; the rats in control group were subjected to +10 Gz exposure for 5 min; the rats in Rd group were administered ginsenoside Rd 30 mg/kg intravenously 30 min before exposure to +10Gz for 5 min; the rats in vehicle group were administered the same volume solvent (20% propylene glycol) intravenously 30 min before exposure to +10Gz for 5 min. The changes of the locomotivity were evaluated by open field test and the ability of learning and memory by water maze.Results In the open field test,the times of rat rears were increased significantly and the time spent in center square was decreased significantly in the Rd group compared to control group (P0.05); there was no significant difference in the times of rears and the time spent in center square between the vehicle group and the control group(P0.05). In the water maze test,the escape latency to find the platform was reduced significantly and the time spent in the probe test was increased significantly in the Rd group compared to control group(P0.05); there was no difference in the escape latency to find the platform and the time spent in the probe test between the vehicle group and the control group(P0.05). Conclusion Ginsenoside Rd can ameliorate the impairment of locomotivity and the ability of learning and memory induced by exposure to +Gz.
