摘  要:目的探讨ERK1/2信号通路在赖氨匹林(aspisol)对人宫颈癌HeLa细胞抑制中的作用。方法分别采用MTT比色法、标准细胞集落形成分析法和Annexin V/PI双染法分析不同浓度aspisol(1、5、10mmol·L-1)对HeLa细胞增殖、细胞集落形成及细胞凋亡的影响;Western blot方法检测细胞外信号调节蛋白激酶(ERK1/2)/磷酸化(P-ERK1/2)和COX-2的表达。结果 aspisol(1、5、10mmol·L-1)可呈浓度依赖性抑制HeLa细胞的生长、降低克隆形成数量、诱导HeLa细胞的早期凋亡率、下调P-ERK1/2及COX-2的表达水平(P<0.01),但不影响总ERK表达(P>0.05)。结论 aspisol对宫颈癌HeLa细胞有抑制增殖、诱导凋亡的作用,其作用机制可能与抑制ERK1/2的活化进而下调COX-2的表达有关。Aim To explore the role of ERK1/2 MAPK signaling transduction pathway in the inhibition of human cervical cancer HeLa cells by aspisol.Methods The effects of aspisol at 1,5,10 mmol·L-1 on cell proliferation,colony formation,early apoptosis rate of HeLa cell were detected by MTT assay,Standard Colony-forming assay,flow cytometry respectively.The ERK1/2,P-ERK1/2,COX-2 protein expressions were detected by western blot.Results The growth of HeLa cells was inhibited;clony numbers were decreased;early apoptotic rates were deduced and expressions of P-ERK1/2 protein and COX-2 protein in HeLa cells were down-regulated by aspisol at 1,5,10 mmol·L-1(P 0.01) and its inhibition effect was related with time and dose of drug,but it did not affect total ERK1/2 expression(P 0.05).Conclusion Aspisol can inhibit cervical cancer HeLa cells proliferation and induce apoptosis.Its mechanisms may be involved in inhibiting ERK1/2 activation and down-regulating the expression of COX-2 protein.
