摘  要:目的通过与聚乳酸(polylactic acid,PLA)进行对比,了解聚丁二酸丁二醇类聚酯[poly(butyl succinate),PBS]/聚乳酸(PLA)合金材料植入动物体内后组织相容性情况。方法选取60只SPF级SD大鼠,按随机数字表法分为4组:Sham组、PBS/PLA组、PBS组和PLA组,每组15只。每只大鼠背部做1个切口,PBS组植入单纯PBS材料,PBS/PLA组植入PBS/PLA合金材料,PLA组植入PLA材料,Sham组切开皮肤后立即缝合,不做其他处理。植入后1、4、8、12、16周后,实验组切取包裹材料的皮下组织,对照组切取切口处皮下组织。主要观察指标:观察切口大体表现;HE染色后光学显微镜观察组织中性粒细胞、淋巴细胞浸润程度和毛细血管密度;Masson染色观察材料周围纤维包裹程度;TUNEL法光镜观察材料对皮下组织凋亡情况。结果第1、4、8、12、16周结果提示PBS/PLA组与PBS组和PLA组相比,3组材料引起的局部炎性细胞浸润和淋巴细胞相似,组织急慢性炎症反应无明显差异;纤维包裹程度PBS/PLA组介于PBS组和PLA组之间(P<0.05);对于材料引起的局部组织细胞凋亡反应3组间也未见明显差异(P>0.05)。结论 PBS/PLA合金材料与PLA材料相比组织相容性未见明显差异;PBS/PLA合金材料具有较好的局部组织相容性。Objective To investigate the in vivo sternal fixation of poly(butyl succinate)(PBS)and polylactic acid(PLA)alloy material by comparing its biocompatibility of polylactic acid(PLA).MethodsSixty SD rats were randomized into 4 groups(n=15).PBS,PLA and PBS/PLA alloy were implanted subcutaneously in the corresponding rat dorsum respectively.Sham group was designed as the control group without any material implantation.In 1,4,8,12 and 16 weeks after implantation,the implanted materials and the surrounding tissues were extracted from the rats,with 3 animal of each time points.The collected implants and surrounding tissue samples were sectioned and stained with hematoxylin and eosin and Masson for histological changes.In addition,the terminal deoxynucleotidyl transferase(TdT)-mediated UTP nick end labeling(TUNEL)apoptosis assay were used for assessment of the subcutaneous tissue apoptosis.ResultsThe PBS/PLA alloy group showed no obvious difference in local acute and chronic inflammation response in contrast with PLA and PBS groups.The thickness of the fiber capsule surrounding the implants in PBS/PLA alloy group was between that of PLA group and PBS group(P〈0.05).The tissue apoptosis in the 3 groups demonstrated no obvious difference statistically(P〉0.05).ConclusionIn contrast with PLA,the PBS/PLA alloy has an acceptable sound in vivo biocompatibility.
