摘  要:目的建立五鹤续断中熊果酸的含量测定方法,并以此作为五鹤续断质量控制指标之一。方法采用高效液相色谱法,ReliasilC18柱(250mm×4.6mm,5μm);流动相为甲醇-水-三乙胺(90:10:0.03,冰乙酸调节pH为6.0);检测波长210nm;柱温23℃。结果熊果酸保留时间为12.35min,进样浓度在23.8~714.0μg·mL-1线性关系良好(r=0.9999),样品平均回收率为98.73%,RSD=0.51%。结论该方法简便易行、灵敏、准确,可用于五鹤续断中熊果酸的含量测定。Objective To establish a method for determination of ursolic acid as a triterpene in Dipsacus asper Wall.,and which is used as the quality control of Dipsacus asper Wall.Methods The Reliasil C18 was used in the HPLC method and mobile phase was composed as methanol-water-triethylamine(90:10:0.03,pH=6.0 adjusted by acetic acid) with detection wavelength at 210 nm and with column temperature at 23 ℃.Results Retention time was 12.35 min,the linear ranges of ursolic acid was 23.8~714.0 μg·mL-1 and the average recovery of ursolic acid was 98.73 %,RSD=0.51%.Conclusion This method is simple,susceptible,accurate,and suitable for the determination of ursolic acid in Dipsacus asper Wall.
