摘 要:目的建立痕量维生素B1的双水相萃取与荧光测定。方法用钼酸钠作为维生素B1的氧化剂,取适量氧化后的硫胺荧加入已盛有一定量聚乙二醇6000、柠檬酸三钠和缓冲液的分液漏斗中,振荡静置后分离上下相,在最佳激发、发射波长(310nm、441nm)处测定有机相的荧光强度。结果在4.0×10-4~2.0×10-2μg/ml的线性范围内,维生素B1的回归方程为y=52.79+21.86x,r=0.9992,该方法检出限为0.46ng/ml,回收率为86.0%~107.2%,相对标准偏差为1.92%~2.66%。结论该方法具有操作简单,准确、灵敏的特点,适用于痕量维生素B1的测定。Objective To establish a new way for the determination of trace vitamin B1 by fluorimetry after aqueous two phase extraction.Methods Sodium molybdate was used as the oxidants of vitamin B1,taking appropriate oxidated thiamine to the separating funnel which was filled with PEG 6000,sodium citrate and buffer after oscillation,put it aside,separating the upper and lower phase,determined the fluorescence of organic phase at the optimum wavelength of 310 nm/441 nm.Results Under the optimal conditions,the extraction method presented linear dynamic ranges of 4.0 ×10^-4-2.0×10^-2 μg/ml,regression equation:y= 52.79+21.86x,with limits of detection of 0.46 ng/ml.The rates of recovery were 86.0%-107.2% and the RSDs were1.92%-2.66%.Conclusion Compared with the other methods,this method is simple,sensitive,accurate and applicable to the determination of trace vitamin B1.
