摘  要:取新生1d的ICR小鼠的脑,经过机械分离和体外培养获取神经干细胞。利用免疫组化和RT-PCR对神经干细胞进行鉴定,发现原代的神经干细胞的纯度能达到95%以上。通过计数绘制第1代和第4代神经干细胞生长曲线图,发现二者生长曲线非常相似,说明第4代的神经干细胞仍具有较强的生长能力。在体外利用聚合后0.01μmol/L、0.1μmol/L、1μmol/L、2.5μmol/L的Aβ1-42寡聚体分别作用于神经干细胞8 h、24 h和48 h,通过MTT和克隆球计数法检测Aβ1-42对神经干细胞的增殖的影响,结果表明,随着Aβ1-42浓度增加和作用时间的延长,细胞的存活率逐渐降低。通过双盲法,免疫荧光下统计Aβ1-42寡聚体作用后神经干细胞的转分化率,检测Aβ1-42对神经干细胞的分化的影响,发现Aβ1-42对神经干细胞的分化没有明显影响,且对Aβ1-42浓度和作用时间也无依赖性。Primary neural stem cells culture were prepared from 1 day neonatal ICR mouse.The brain tissue was mechanically separated,and neural stem cells were isolated and obtained.The purity of primary cell is more than 95% after identifying by immunofluorescence and RT-PCR.It is find that the fourth passage cells were similar to primary neural stem cells by growth curve,which showed the fourth passage neural stem cells still owned strong growth capability.For testing the effects of Aβ on the proliferation of neural stem cells,we exposed neural stem cells to 0.01μmol/L,0.1μmol/L,1μmol/L,2.5μmol/L oligomeric Aβ1-42 for 8 h,24 h and 48 h in vitro respectively.The results indicated the proliferation declined in a dose-and time-dependenc.For testing the effects of Aβ1-42 on the differentiation of neural stem cells,we made a statistic of trans-differentiation percent by double blind method,which showed Aβ1-42 had not obvious effection to neural stem cells' differentiation in a dose-and time-dependence.
