摘  要:目的探讨经腹侧面暴露SD大鼠下丘脑的手术方法,为电损毁、核团内微量给药、神经元单位放电记录等技术用于下丘脑研究提供方便。方法通过剪开25只SD大鼠下颌骨、向尾侧部分牵拉舌头、切开软腭、用牙钻在基蝶骨钻孔等步骤暴露大鼠下丘脑,并通过放电特点鉴定神经元活性。结果用玻璃微电极记录到下丘脑弓状核(ARC)神经元的单位放电。结论经腹侧面暴露大鼠下丘脑的手术操作简便,成功率高,能方便地暴露大脑腹侧面的核团(如弓状核和正中隆起),以便后续研究。Objective To explore the ventral surgical approach to the rat hypothalamus as the convenient experimental methods for the study of hypothalamus(such as electrical lesioning,intranuclear microinjection and neuronal discharge recording).Methods The ventral surgical approach to the Sprague-Dawley rat hypothalamus was performed by retracting the hemisectioned mandibles laterally,drawing the tongue caudally,dividing the soft palate and removing the sphenoid bone with a dental drill etc.The neuronal activity could be indentified by discharge characteristics.Results The unit discharge was recorded extracellularly with a glass microelectrode from arcuate nucleus.Conclusion The ventral surgical method is simple and highly efficient.By this technique,neural structures at the base of the brain(such as the arcuate nucleus and the median eminence) are easily exposed and thus become accessible to direct inspection under a dissecting microscope.
