摘 要:目的探讨老龄大鼠脑内M胆碱受体和β肾上腺素受体的变化规律,以及不同补肾中药的调整作用。方法以3H-QNB和3H-DHA为标记配基,分别对M胆碱受体和β肾上腺素受体作放射配基结合分析。采用单点法测定受体数(RT)。结果早期衰老大鼠(22~26月龄)脑M受体的RT值降低约8%,滋阴药知母生地上调M受体的作用有显著意义,β受体的密度无显著变化,5种中药配方也均无明显作用;原期衰老大鼠(30~32月龄)脑内M受体RT值降低幅度约为10%左右;β受体RT值则显著大幅度下降,平均降低20%以上,补气药黄芪在上调M受体的同时,也明显上调β受体,其它4种中药对M和β受体作用均不显著。结论衰老早期以M受体下降为主,滋阴药知母生地有效康老晚期除M受体继续降低外,β受体降低幅度更大,补气药黄芪有效。Objcctivc To investigate the change of muscarinic receptor (MR) and β-adren0ceptors in the brain of aged rats and the effect of five different tonic - drugs on abovereceptors. Methods The densities (RT) of MR and β- adrenoreceptors were determined byradioligand binding assay with 3H - QNB and 3H - DHA, respectively. It was used a singleradioligand dose of saturation area. Results The RT values of brain MR of early aged rats(22~26 months) were significantly lower than those of young rats. The decrease was about8%. Parallel experiments revealed that when the above rats were treated by the Yin - tonics(SZ), the SZ have a significantly up - regulatory effect on MR densities- The RT values ofβ- adrenoreceptors were no difference between the early aged rats (22~26 months) andyoung rats, and also were not altered by treatment with five drugs. The RT values of MR ofthe later aged rats(3O~32months) were decreased ab0ut 10 % , but β - adrenoreceptor densitieswere significantly decreased (more than 2O%). The densities both muscarinic receptors andβ- adrenoreceptors were up - regulated significantly with treatment of Qi - tonics.ConcIusions The results dem0nstrated that the densities of MR of early aged rats weremainly reduced and were increased in aged rats, treated with SZ. For later aged rats,the densities of MR continued reducing, and β - adrenoreceptors were significantly reduced.Qi - tonics has a significantly up - regulatory effect on these above receptors.
