摘 要:目的探查检测血吸虫循环抗原多种特异组分表位的高效探针系列。方法采用双位点夹心酶联试验对捕获抗体与检测抗体的最佳组合进行了同相或异相的配伍测试。结果经过筛查,获得了分别能高效检测血吸虫优势循环抗原肠相关CAA、CCA及一个卵相关SEA组分的若干同相和异相配伍组合,其中一组异相配伍可同时高效测出CAA和CCA组分,最高检测极限对这三种表位均可达1ng/ml水平。结论提示此三种组合中最易检示的机率在CAA组分,而CCA的组合机率相对较少。在异相组合中检测的特异性似取决于捕获抗体的质量和同型(isotype)性质。0hjective To search highly efficient probe systems for specific detections ofSchistosoma- circulating antigen (CAg) epitope of various fractions. Methods Optimalmatching trails were investigated between capture - and detecting- monoclonal antibodies(McAbs) in two- site sandwich ELISAs by homologous or heterologous com-binations. Results Through intensive matching, several sensitive detecting systems eitherin hemologous or heterologous combinati0ns were worked out specifically for the majorschistosomal CAg - components, gut - associated CAA or CCA as well as for one of theegg - associated SEA circulating fractions. Among these effective combinations, oneheterologous combination could detect both CAA and CCA simultaneously being moreeffectively for the CCA fraction. The highest detecting limit for the 3 kind of epitopesreached to a level as high as equal to or less than 1ng/ml. ConcIuhon The results indicatedthat CAA was the component most readily to be detected among the three, whereas CCA wascomparatively more difficult. lt is suggested that in heterologous system, detectingspecificity might be more associated with the quality and the isotype of the capture - McAbused, while detecting sensitivity was closely related to the RZ value of the batch of thelabeled McAb conjugate.
