摘 要:目的探讨不同尿酸浓度对DOCA-salt高血压大鼠内皮细胞的影响。方法根据Theodora Szasz的方法制备DOCA-Salt高血压大鼠模型,4周后与对照组分别培养胸主动脉内皮细胞,然后用10、50、100μm尿酸和载体NaOH和内皮细胞孵育1/2h,比较ET-1、NO及NF-κBmRNA表达的差别。结果在100μm尿酸的作用下,DOCA-salt高血压大鼠组血压ET-1和NF-κBmRNA表达均明显高于对照组(P<0.05),而NO的表达明显低于对照组(P<0.05)。结论 100μm尿酸促进了DOCA-salt高血压大鼠内皮细胞功能的失调,可能通过NF-κB影响了ET-1和NO的表达,从而促进并加重高血压及其血管的病变。Objective To explore the effect of different concentration of uric acid on endothelial cells in DOCA--Salt Induced Hypertensive Rats.Methods We were prepared DOCA--Salt hypertensive rats according to Theodora Szasz's method. After 4 weeks, we cultured aortic endothelial ceils of two groups,and then it were incubated 1/2h with 10 μ M,50 μM,100 μM uric acid and NaOH to compare the difference in concentration of NO. ET^-1and the expression of NF-κB mRNA.Results Uric acid(100 M)was significantly alter blood pressure,and the concentration of NO. ET-land the expression of NF-κ B mRNA between these two groups(P〈0.05).Conclusion Uric acid(100 μM) promoted function disorder of endothelial cell in DOCA-Salt induced hypertensive rats,NF-- ~ B might affect the expression of ET--land NO on endothelial cells,might contribute to and aggravate hypertension and vascular disease.
