活性维生素D_3对大鼠系膜增生性肾炎细胞增殖与凋亡的影响

摘　　要：目的:研究活性维生素D_3对系膜增生性肾小球肾炎细胞增殖与凋亡的影响。方法:72只雄性SD大鼠随机分为对照组、肾炎组、活性维生素D_3干预组,每组24只,分别于造模和给药后第1,3,7,14天每组随机处死6只大鼠,肾组织标本HE染色确定肾脏病理损害分级,免疫组化法检测PCNA、Bcl-2在肾组织中的分布,Western blot检测PCNA蛋白的表达量。结果:与对照组相比,肾炎组PCNA和Bcl-2表达明显增强(P<0.01);活性维生素D_3干预组PCNA和Bcl-2表达明显减少(P<0.01)。PCNA、Bcl-2表达与病理分级呈正相关(P<0.01),PCNA、Bcl-2各指标间的表达呈正相关(P<0.01)。结论:活性维生素D_3可以通过抑制PCNA的表达而抑制系膜细胞增生,减少Bcl-2表达促进凋亡,延缓疾病的进展。Objective ： To study the role and mechanism of active vitamin D in mesangial proliferative golmerulonephritis. Method： Seventy-two male SD rat were devided into three average groups randomly：saline control rats, nephritis rats and active vitamin D3 rats. Executed 6 rats of each group randomly and collected the renal tissue samples at day 1,3,7 and 14 ,respectively. ①The pathologic abnormalities of MsPGN was observed by HE dyeing;② The expression of PCNA and Bel-2 were detected by immune-histochemistry. ③ The expression of PCNA protein was detected by western blot in each group. Result： Compared to control group, the expression of PC- NA and Bcl-2 in glomerulo- nephritis group was significantly increased （P 〈 0. 01 ）, respectively; compared to nephritis group, the expression of PCNA and Bcl-2 in active vitamin D3 group was significantly reduced （ P 〈 0. 01 ）. The expression of PCNA and Bcl-2 be- tween pathologic classify was positive correlation （ P 〈 0. 01 ） , respectively. The expression of PCNA, PCNAmRNA and Bcl-2 were posi- tive correlation with each other （P 〈 0. 01 ）. Conclusion： Active vitamin D3 can suppress the proliferation of mesangial cells by inhibi- ting the expression of PCNA and PCNAmRNA,promote apoptosis by reducing the expression of Bcl-2, attenuates progression of the mesangial proliferative glomerulonephrisis.
