摘  要:应用两种属于同一分类的硝酸银(或酸式)银染方法检测聚丙烯酰胺凝胶电泳分离后的不同的蛋白质,发现成份相同的凝胶产生了深浅不同的背景,从而揭示了凝胶背景产生的另一个原因是与所使用的银染方法中的化学条件有关.这两种方法的监测灵敏度相同,都可检测纳克级的微量蛋白质.同时,对银染方法使用中出现的异常情况进行了分析.Two different silver staining methods, silver nitrate or acidic methods, were employed to detect proteins in sodium dodecyl sulphate-polyacrylamide gels following the electrophoresis of a variety of protein samples. Differences in background intensity of silver stained gels whose components were identical suggested that the background formation was due to the conditions used in specific silver staining method. The two methods were found to have same sensitivity to detect protein in single nanogram quantity. Some unusual silver staining characteristics were analyzed.
