摘  要:目的研究MMP-9和CC10在小鼠变应性鼻炎中的表达和两者之间是否有相关性。方法用卵清蛋白来建立野生型小鼠和CC10基因敲除小鼠变应性鼻炎动物模型。分别用免疫组织化学、过碘酸-希夫染色、苏木素-伊红染色,检测小鼠鼻黏膜组织CC10和MPP9表达、杯状细胞、浸润的嗜酸性粒细胞。结果和对照组相比,MMP-9、嗜酸性粒细胞、杯状细胞,在变应性鼻炎小鼠鼻黏膜中表达明显升高,并且在CC10基因敲除小鼠中升高更加显著,均P<0.01。相反,而和对照小鼠相比,CC10在变应性鼻炎小鼠鼻黏膜中表达明显下降,P<0.01。MMP-9和CC10表达呈显著负相关,相关系数为-0.57,P<0.01。结论 MMP-9和CC10参与了小鼠变应性鼻炎的发病过程,而且CC10直接或者间接抑制了MMP-9的表达。Objective To investigate the expression of MMP-9 and CC10 and its correlation in allergic rhinitis model of mice.Methods A allergic rhinitis model was established by ovalbumin injection in wide type and CC10 gene knockout mice.The changes of eosinophilic cells,and goblet cells,and the expression of MMP-9 and CC10 in nasal mucosa were examined by hematoxylin and eosin staining,periodic acid-schiff staining and immunohistochemistry methods,respectively.Results compared with control group,eosinophilic cells,and goblet cells,and the expression of MMP-9 were significantly increased in allergic rhinitis model group(P0.01),and the increases were more obviously in CC10 gene knockout allergic rhinitis model group(P0.01),However,CC10 protein expression in nasal mucosa of allergic rhinitis model group was significantly decreased as c ompared to control group(P0.01).Moreover,the expression of MMP-9 was significantly negatively correlated with CC10 protein expression(r=0.57,P0.01).Conclusion MMP9 and CC10 might play crucial role in the pathogenesis of allergic rhinitis of mice and CC10 may directly or indirectly inhibit the expression of MMP-9 in allergic rhinitis.
