摘 要:目的研究α-硫辛酸对体外培养兔晶状体氧化损伤的保护作用。方法将正常兔晶体分为对照组,氧化损伤组和不同浓度α-硫辛酸保护组,培养48 h后观察各组晶状体混浊情况,测定各组晶状体谷胱甘肽(GSH)及丙二醛(MDA)含量及总抗氧化能力(T-AOC)。结果氧化损伤组晶状体全部混浊,对照组基本透明、0.5 mol/L和1 mol/Lα-硫辛酸组分别为67%和33%。和对照组相比,氧化损伤组T-AOC及GSH含量明显降低,MDA含量明显升高,0.5 mol/L和1 mol/Lα-硫辛酸组上述指标有不同程度改善。结论α-硫辛酸可保护晶状体免受氧化损伤,减少白内障的发生。1 mol/Lα-硫辛酸效果最佳。Objective To evaluate the effect of α - lipoie acid on protecting lens against oxidative stress. Methods Rabbit's lenses cultured in vitro were divided into 4 groups :control group , oxidative stress group,0.5 mol/L and 1 moL/L α - lipoic acid treatment groups. The transparency of all lenses was examined after above lenses incubated for 48 h. At the same time, the levels of T - AOC , GSH and MDA were measured respectively. Results After incubation for 48 h , all lenses of oxidative stress group developed into cataract ;67 % lenses of 0.5 mol/L α - lipoic acid group and 33 % lenses of 1 nlol/L α - lipoic acid group were showed swelling. The level of MDA was increased and levels of T - AOC and GSHwere decreased significantly in oxidative stress group compared with those of tbem in control group ( P 〈 0.01 ). The level of MDA was significantly decreased but the levels of other indexes were increased in 0.5 mol/L and 1 mol/L α - lipoic acid groups compared with those of them in oxidative stress group. Conclusion α - lipoic acid can protect lens against oxidative stress ,preventing it from lipid peroxidation and cataract formation in vitro. 1 mol/L α- lipoic acid may be more beneficial.
