摘  要:目的探讨灭幽汤对幽门螺杆菌(H.pylori)感染小鼠胃黏膜组织核转录因子-κB(NF-κB)和核转录因子-κB抑制蛋白(IκBα)表达的影响及其作用机制。方法采用H.pylori菌液灌胃法造模,将造模成功的小鼠按体重分为模型组、灭幽汤组、三九胃泰组、丽珠胃三联组,除模型组每日予生理盐水外,其余组分别予灭幽汤、三九胃泰、丽珠胃三联给药。采用免疫组化法观察胃黏膜组织NF-κB和IκBα蛋白的表达情况。HE染色观察小鼠胃黏膜的病理组织改变。结果正常对照组胃黏膜细胞胞浆有少量NF-κB蛋白表达,胞核内无表达,而模型组NF-κB主要位于细胞核内。与模型组比较,灭幽汤组明显抑制胃组织细胞内NF-κB的核移位,灭幽汤组IκBα的磷酸化程度较模型组明显降低(P<0.05)。结论灭幽汤可能通过对降低IκBα的磷酸化程度,抑制NF-κB蛋白表达,发挥其治疗作用。Objective To study the effect of Mieyoutang on NF-κB and IκBα expressions in Helicobacter pyloriassociated gastritis mucosa rats. Methods With H.pylori bacilli gavage method of modeling, the successful model were randomly divided into model group, Mieyoutang high-dose group and Lizhuwei group. NF-κB and IκBa protein expressions were observed by immunohistochemical staining gastric mucosa. HE staining was used to observe the pathological changes of gastric mucosa in mice. Results In normal gastric mucosa cells of control group, there was a small amount of NF-κB protein expression, no expression within the nucleus, while NF-κB mainly located in the nucleus in model group. Compared with model group, Mieyoutang significantly inhibited NF-κB nuclear translocation of gastric mucosa cells, and the phosphorylation level of IκBa of Mieyoutang group was significantly lower than the model group (P〈 0.05). Conclusion Mieyoutang may reduce the degree of IκBo phosphorylation and inhibit NF-I〈B protein expression, so as to play its therapeutic effect.
