摘 要:目的观察脂脉宁药物血清对香烟提取物(CSE)诱导的血管内皮(VEC)-304细胞损伤的影响。方法将培养细胞分为CSE组、空白组、脂脉宁大剂量组、脂脉宁小剂量组。采用MTT、免疫组化和TUNEL等方法,观察各组细胞存活率和凋亡率、相关凋亡蛋白表达及氧化反应指标的变化。结果 CSE组存活率最低,凋亡率最高,Wp53、p21、Bax、Caspase-3表达均明显升高,Bcl-2、mp53表达降低,超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)活性、谷胱甘肽(GSH)含量均降低,丙二醛(MDA)含量及一氧化氮合酶(NOS)活性升高,与空白组比较差异有统计学意义;脂脉宁大、小剂量组存活率明显升高,凋亡率降低,Wp53、p21、Bax、Caspase-3表达下调,Bcl-2、mp53表达上调;SOD、GSH-Px、T-AOC活性及GSH含量均升高,MDA含量及NOS活性降低,而脂脉宁大剂量组更明显。结论 CSE诱导的VEC-304细胞凋亡与其过氧化反应密切相关,脂脉宁有抑制细胞凋亡及抗氧化作用。Objective To approach the effect of Zhimaining drug serum on cigarettes extract (CSE) induced vessel endothelium (VEC)-304 cell demage. Method The survival rate, apoptosis rate, apoptosis protein expression and oxidation index of blank group, CSE group, Zhimaining large dose group and Zhimaining small dose group of cultivated were detected by method of MTr, TUNEL, immunohistochemistry and so on. Results The cell survival rate of CSE group was the lowest and apoptosis rate was the highest. The expression of Wp53, p21, Bax, Caspase-3 were obviously higher and expression of Bcl-2, mp53 were lower than blank group. The content of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), totol antioxygen capability (T-AOC), and glutathione (GSH) were lower and malondialdehyde (MDA), nitrogen monoxidum enzyme (NOS) were higher than blank group. In Zhimaining large and small dose group, the cell survival rate was higher and apoptosis rate was lower, Wp53, p21, Box, Caspase-3 expression down-regulation, and Bcl-2, mp53 expression up-regulation, SOD, GSH-Px, T-AOC activity, GSH content were higher, and MDA content, NOS activity were lower than blank group. Conclusion The CSE induced VEC-304 apoptosis is intimately correlated to peroxidation reaction.
