摘　　要：目的探讨凋亡诱导因子(AIF)在铜绿假单胞菌(PA)上清液诱导J774细胞凋亡中的表达。方法选用J774细胞作为PA上清液感染的体外细胞模型,使细胞与15%的PA上清液混合,37℃水浴中培养3、6、9 h,采用G im sa染色技术、荧光染色技术、流式双染技术进行J774细胞凋亡检测,W estern b lot法测定AIF含量。结果 PA上清液与细胞培养3 h即有少数J774细胞发生凋亡,随时间延长凋亡细胞数量逐渐增多;PA上清液刺激3 h后AIF开始出现低量表达,6、9 h时AIF表达更加明显。结论 PA上清液可诱导J774巨噬细胞凋亡,其作用具有时间依赖性;AIF蛋白与PA上清液诱导巨噬细胞凋亡明显相关。Objective To explore the expression of apoptosis-inducing factor（AIF） protein in the apoptosis of J774 cells induced by the cell-free supernatant from pseudomonas aeruginosa.Methods J774 cells as the model in vitro,by the cell-free supernatant from pseudomonas aeruginosa at a concentration of 15%,after 3,6,9 hours of incubation in 37 ℃ bain-marie,with Giemsa＇s staining,fluorescence staining and AnnexinV-FITC/PI assay,J774 cells apoptosis were detected.With Western blot method AIF levels were detected.Results After 3 hours of incubation,there were a small number of apoptosis of J774 cells.As the time extended,the amount of apoptosis cells increased.After 3 hours of incubation,there was low expression of AIF protein.After 6,9 hours,the expression level of AIF protein enhanced obviously.Conclusions The cell-free supernatant from PA could induce apoptosis of J774 cells with a time-dependently.AIF protein might correlate with PA induced macrophage apoptosis.
