摘 要:目的:建立高效液相色谱法测定人工养殖蟒蛇胆汁中牛磺胆酸钠含量的方法。方法:采用Phe-nomenex Luna C18色谱柱(250mm×4.60mm,5μm);流动相为乙腈-0.15%磷酸氢二钠缓冲盐(pH=3.0)溶液,梯度洗脱;流速为1.0 mL.min-1;检测波长为203nm;柱温为35℃。结果:牛磺胆酸钠在0.0051-0.0816mg.mL-1的浓度范围内线性关系良好,r=0.9997。平均回收率为102.1%,RSD=1.6%(n=6)。7批蟒蛇胆汁中牛磺胆酸钠的平均含量7.35mg.g-1。结论:该方法简便、可行,能准确测定人工养殖蟒蛇胆汁中牛磺胆酸钠的含量,为蟒蛇胆汁的质量控制提供理论依据。Objective: To establish a HPLC method for the determination of sodium taurocholate in the bile of python.Methods: After prepared,the sample was separated at 35℃ on a Phenomenex Luna C18(250mm×4.60mm,5μm) eluted with acetonitrile mixed with 0.15% disodium hydrogen phosphate(add phosphoric acid to pH 3.0) in gradient mode.The detection wavelength was 203nm,and the flow rate was 1.0mL·min-1.Results: The calibration curve was linear within the concentration range 0.0051~0.0816mg·mL-1(r=0.9997),and the average recovery was 102.1%,SD = 1.6%(r=6).The average content of sodium taurocholate in seven batches of the bile of python was 7.35mg·g-1.Conclusion:The method was simple,rapid,precise and good reproductive.It can be use for the determination of sodium taurocholate in the bile of python.
