摘　　要：目的:建立激素性股骨头坏死家兔模型,观察和评价中药对早期激素性股骨头坏死的防治效果,并探讨其作用机理。方法:选用成年雄性日本大耳白兔随机分为3组:正常组、治疗组、模型组。治疗组和模型组经耳缘静脉注射马血清两次,第1次10mL.kg-1,间隔3 w后,第2次注射6mL.kg-1,间隔2w后,连续3d每日1次腹腔注射甲强龙45mg.kg-1。注射激素后第2d治疗组用骨蚀灵胶囊进行灌胃,正常组和模型组两组均以等体积的蒸馏水灌胃。在灌胃第4w和第8w时分批处死动物,迅速取出股骨头,截取股骨头取材后迅速投入液氮中,从骨组织中提取核酸,应用RT-PCR法检测不同时间点Bcl-2 mRNA表达。结果:联合应用马血清和甲强龙的方法可成功造成家兔激素性股骨头坏死模型,成功率高,死亡率低。应用液氮下迅速研磨骨组织的方法成功提取了骨组织核酸,RT-PCR法检测不同时间点Bcl-2 mRNA表达,Bcl-2 mRNA的表达随着时间推移各组表达量逐渐降低,差异有统计学意义(P<0.05)。结论:中药骨蚀灵胶囊通过调节股骨头局部血循环,可以抑制细胞凋亡,同时促进成骨和组织修复,作用全面,通过多种途径防治激素性股骨头坏死早期病理变化,防止病变的进展。通过探讨中药防治激素性股骨头坏死机理的研究,能够加深对激素性股骨头坏死早期病理变化的认识,针对其中的关键环节进行干预,可以更好的预防和治疗糖皮质激素所导致的骨坏死。Objective：Through establishing the rabbit model of steroid-induced avascular necrosis of the femoral head,we observe and investigate the prevention and treatment effect of traditional Chinese medicine ＂Gu Shi Ling＂ capsule for the early-stage steroid-induced avascular necrosis of the femoral head.Methods：We choose the adult Japanese white rabbits,which were randomly and equally divided into normal control group,model group and treatment group.The rabbits in model group and treatment group received horse serum intravenously twice,which received 10ml/kg of horse serum at the first time,6ml/kg were injected at the second time three weeks later,then 45mg/kg methyprednisolone were intraperitoneal injected for three consecutive days at an interval of two weeks.Then from the next day the rabbits of the treatment group began to receive ＂Gu Shi Ling＂ capsule lavage,the other two group received equal volume of distilled water lavage.Rabbits of each group were killed at the 4th,8th week respectively,the two heads of femur were taken out quickly.The heads of femur was put into liquid nitrogen quickly,nucleic acid were extracted from bone tissue,the mRNA expression of Bcl-2 were detected by RT-PCR.Results：The rabbit model of steroid-induced avascular necrosis of the femoral head can be induced successfully by the combined use of horse serum and methyprednisolone,which have the advantage of a high success rate and lower rate of death.Nucleic acid were extracted successfully by grinding bone in liquid nitrogen.The mRNA expression of Bcl-2 were detected by RT-PCR at different point-in-time successfully.The mRNA expression of Bcl-2 decrease gradually as time goes on,there are obviously differences in the three groups（P0.05）.Conclusion： The Chinese medicine ＂Gu Shi Ling＂ capsule plays a comprehensive,thorough role in prevention and treatment for the early-stage pathologic changes of steroid-induced avascular necrosis of the femoral head.It can regulate the blood circulation of the femoral head,inhibit apopt
