重庆金佛山地区东方蜜蜂遗传多样性研究

摘　　要：对重庆金佛山地区31个东方蜜蜂样本的线粒体DNAtRNAleu～COⅡ基因进行了扩增和测序,同时还测定了重庆市其他地区6个东方蜜蜂样本的相同基因序列和从GenBank中下载了中国境内其他地区的10个东方蜜蜂样本相同基因序列,利用相关软件对重庆金佛山地区东方蜜蜂进行了遗传多样性分析.结果表明:金佛山地区东方蜜蜂存在4个单倍型,主体单倍型与国内大部分地区的单倍型相同,其余3个单倍型在国内尚未见报道;聚类分析显示金佛山东方蜜蜂单倍型H2和海南海口东方蜜蜂聚在一起.推测金佛山地区东方蜜蜂与国内大部分地区东方蜜蜂具有相似的基因库,但同时又具有自己独特的遗传背景.The mitochondrial DNA tRNAleu～COⅡgene in 31Apis ceranacolonies from Jinfo Mountain of Chongqing was amplified and sequenced.6Apis ceranacolonies from other places of Chongqing were also determined using the same method.Furthermore,10 mtDNA tRNAleu～COⅡgene sequences ofApis cera-nafrom other areas in China were downloaded from GenBank for comparative analysis.The alignments were performed using related softwares.The results showed that there were 4 haplotypes inApis cerana colonies from Jinfo Mountain of Chongqing,one of them was main haplotype and same as that from many other places in China,the other 3 haplotypes were different from the otherApis ceranain China.By cluster analysis,haplotypeⅡfrom Jinfo Mountain was clustered with theApis ceranafrom Haikou of Hainan Province.Apis ceranafrom Jinfo Mountain of Chongqing has the similar genetic background as theApis ceranafrom other areas in China,meanwhile,some haplotypes are unique.
