对孟鲁司特钠分散片的人体生物等效性研究

摘　　要：目的:比较国产孟鲁司特钠分散片(受试制剂)与进口孟鲁司特钠片(参比制剂)的人体生物利用度。方法:20名健康受试者进行随机交叉试验,分别先后单次口服10mg两种孟鲁司特钠制剂,采用LC-MS法测定血浆中孟鲁司特浓度。结果:受试制剂和参比制剂的主要药动学参数分别为:t1/2(4.6±0.5)和(4.6±0.5)h,tmax(3.8±1.1)和(3.9±1.0)h,cmax(408.6±79.1)和(498.7±114.2)ng/ml,AUC0～24(3541±643.7)和(3143±436.8)ng.h.ml-1,AUC0～∞(3655±660.7)和(3260±450.4)ng.h.ml-1,结果表明主要药动学参数无显著性差异,受试制剂的相对生物利用度为(90.6±16.1)%。结论:国产孟鲁司特钠分散片与进口孟鲁司特钠片在健康人体内生物等效。Objective：To compare the bioequivalence of domestic montelukast sodium dispersible tablets and imported montelukast sodium tablets in human body.Methods：A single oral dose of 10 mg test or reference montelukast was given to 20 volunteers in a randomized cross-over study,and the plasma concentration of montelukast was determined by liquid chromatography-tandem mass spectrometry（LC-MS）.Results：The main pharmacokinetic parameters of test and reference preparations were as follows：t1/2（4.6±0.5） and（4.6±0.5） h,tmax（3.8±1.1） and（3.9±1.0） h,cmax（408.6±79.1） and（498.7±114.2） ng/ml,AUC0-24（3 541±643.7） and（3 143±436.8） ng.h.ml-1,AUC0-∞（3 655±660.7） and（3 260±450.4） ng.h.ml-1,respectively.There were no significant differences in main pharmacokinetic parameters between the two preparations,and the relative bioavailability of test preparation was（90.6±16.1）%.Conclusion：The domestic montelukast sodium dispersible tablets and imported montelukast sodium tablets are bioequivalent in healthy human.
