摘  要:[目的]对影响魔芋细胞悬浮培养及其次生代谢物葡甘露聚糖产生的主要影响因子进行研究。[方法]利用3,5-二硝基水杨酸测定葡甘露聚糖含量。[结果]在魔芋细胞悬浮液体培养葡甘露聚糖的过程中,采用MS培养基为基本培养基,pH值为6.0时有利于葡甘露聚糖的合成,25g/L乳糖可作为最适碳源;2,4-D在1.0mg/L时葡甘聚糖的积累效果明显;细胞分裂素6-BA使葡甘露聚糖积累明显高于KT,且1.5mg/L时葡甘露聚糖积累量较高,培养15d左右葡甘露聚糖含量达到最大。[结论]MS+2,4-D1.0mg/L+6-BA1.5mg/L+乳糖25g/L,pH6.0,为魔芋细胞悬浮培养生产葡甘聚糖的较优培养基。[Objective] Some major factors affected the cell suspension culture of Amorphophallus and the production of konjac glucomannan were studied.[Method] 3,5-dinitrosalicylic acid method was used to measure the content of KGM.[Result] The optimal media was MS media.The concentration of KGM was the highest on 15th day.The proper pH value was 6.0 and 25 g/L lactose was the better carbon source.The glucomannan accumulation treated with 6-BA was significantly higher than that treated with KT,and the maximal production of glucomannan was obtained when treated with 1.5 mg/L 6-BA.The accumulation of glucomannan was the highest treated with 1.0 mg/L,2,4-D.[Conclusion] MS + 2,4-D 1.0 mg/L + 6-BA 1.5 mg/L + lactose 25 g/L,pH 6.0 gave the best result for glucomannan accumulation.
