丹参酮ⅡA对椎间盘突出症大鼠脊髓背角降钙素基因相关肽表达的影响

摘　　要：目的研究丹参酮ⅡA腹腔注射对椎间盘突出症大鼠降钙素基因相关肽(CGRP)表达的影响。方法 30只雄性SD大鼠,随机分为3组,每组10只:对照组(C组)、椎间盘突出症组(B组)和椎间盘突出症+丹参酮ⅡA组(A组)。A、B两组均硬膜外植入尾椎髓核,C组实施假手术。术后第8天开始注药治疗,A组每日腹腔注入丹参酮ⅡA磷酸钠注射液25mg/kg,B组每日注入生理盐水2mL/kg,C组不做处理。在手术前及注药后5、10、15d分别测定各组大鼠双后肢足底对VonFrey触针诱发的回缩阈值。连续用药15d后,第16天取L5L6段脊髓背角,采用免疫组织化学染色方法观察丹参酮ⅡA对脊髓背角细胞肽类神经递质CGRP表达的影响。结果 A、B组在术后第5～15天内对机械刺激均产生明显的痛觉过敏,用药15d后,A、B组与C组相比,脊髓背角CGRP阳性表达显著增加(P<0.05),A组脊髓背角CGRP阳性表达与B组相比降低,差异有统计学意义(P<0.05)。结论腹腔注射丹参酮ⅡA可降低椎间盘突出症大鼠脊髓背角CGRP表达。Objective To observe the effects of tanshinone ⅡA injected into abdominal space of rats on the expressions of calcitonin gene related peptide in L5-6 dorsal horn neurous of spinal cord.Methods Thirty adult male SD rats weighing 280-320g were randomly divided into 3 groups （n=10 each）：group C contol;group B intervertebral disc herniation experiment and group A intervertebral disc herniation＋ tanshinone ⅡA experiment.The nucle pulposus of coccygeal vertebra were added into epidural space in group A and B,group C were not administered by real operation.Tanshinone ⅡA was administered by injecting into abdominal space once per day for 15 days according to group C.Before operation and after drug injection the posterior paw withdrawal mechanical threshold was measured by Von Frey hairs and the expressions of calcitonin gene related peptide in L5-6 dorsal horn neurous of spinal cord were detected with immunohistochemistry tecmique.Results After operation in 5-15 d all rats of group A and B had significant hyperalgia to mechanical stimulation compared with that of group C（P0.05）,and the expressions of calcitonin gene related peptide in dorsal horn neurous of spinal cord increased in group A and B respectively （P0.05）,but the one in group A was reverse compared with group B （P0.05）.Conclusion The expressions of calcitonin gene related peptide in dorsal horn neurous of spinal cord decrease by injecting tanshinone ⅡA into abdominal space during intervertebral disc herniation in rats.
