摘  要:目的观察苦参碱微乳制剂对小鼠四氯化碳(CCl4)肝损伤模型的保护作用。方法采用CCl4造成小鼠急性肝损伤模型,预防性经皮肤给苦参碱微乳制剂高、中、低剂量(200,100,50mg/kg)和灌胃给苦参碱水溶液(200mg/kg),测定血清丙氨酸转移酶(ALT),天门冬氨酸氨基转移酶(AST)。结果与CCl4组相比,微乳高、中剂量和苦参碱口服水溶液组,ALT均显著降低,微乳高剂量和苦参碱口服水溶液组,AST均显著降低,苦参碱微乳制剂组ALT,AST水平与苦参碱口服水溶液组间无显著差异。结论苦参碱微乳制剂对小鼠急性CCl4肝损伤有降转氨酶作用,具有一定的新药开发前景。Objective To investigate the protective effects of matrine microemulsion on mouse model of liver injury induced by carbon tetrachloride. Methods The mouse model of liver injury was induced by CCl4. The matrine microemulsion by transdermal delivery in high,medium and low-dose ( 200,100,50 mg/kg) ,and water solution of matrine by oral administration( gavage control 200 mg/kg) ,and the activities of alanine transferase ( ALT) and aspartate amino transferase ( AST) in serum were determined. Results Compared with CCl4 group,ALT was significantly decrcased in the groups of high and medium doses of the matrine microemulsion and the group of water solution of matrine,and the AST was significantly reduced in the group of high doses of the matrine microemulsion and the group of water solution of matrine. The ALT and AST levels were no significantly different between the two groups of matrine microemulsion and water solution of matrine. Conclusion Matrine microemulsion plays a role in prevention of acute CCl4 liver injury in mice with reduced transaminase. Matrine microemulsion has a certain prospect in new drug development.
