摘 要:目的观察桃核承气汤对庆大霉素所致大鼠肾毒性损伤模型的治疗作用及部分机制。方法采用腹腔注射庆大霉素法制作大鼠肾毒性模型,通过生化、病理检测和蛋白免疫印迹方法观察各组大鼠肾功能、肾组织病理形态、氧化产物及酶类的变化及信号通路分子p38、核因子κB(NF-κB)蛋白表达的变化,明确桃核承气汤的疗效。结果桃核承气汤治疗一周后,可改善肾功能,减轻肾组织病理改变,显著降低肾皮质MDA含量和NOS酶活性、提高SOD酶活性,NF-κB蛋白表达明显减少。结论桃核承气汤可以通过改善肾脏氧化损伤对庆大霉素肾毒性起到一定的治疗作用。Objective To study the therapeutic effect of Taohe Chengqi Decoction ( TCD) on gentamicin-induced kidney lesion in rats. Methods The renal injury model was established by Gentamicin i. p. The kidney index,pathomorphology,and level of oxidative production and expression of Nuclear Factor-κB ( NF-κB) protein in kidney cortex were measured to observe the effect of TCD on kidney injury of rats. Results After one week therapy,parameters of kidney indexes,oxidative production MDA and enzyme activity of NOS in kidney cortex of Gentamicin group were significantly decreased ( P 0. 05) ,and enzyme activity of SOD was significantly increased( P 0. 05) . The protein expression of NF-κB was significantly down-regulated ( P 0. 05) . Conclusion Taohe Chengqi Decoction has some therapeutic effect on Gentamicin-induced kidney lesion through alleviating renal oxidation.
