结核病患者血清蛋白质/多肽谱的初步研究

摘　　要：目的了解结核病患者血清蛋白质/多肽的表达,为新型结核病血清学诊断方法的研究提供基础依据。方法 (1)应用WCX蛋白质芯片和表面增强激光解析-电离飞行时间质谱(SELDI-TOF-MS)技术对99例肺结核患者和18例肺外结核患者进行了血清蛋白质/多肽谱检测,采用ClinProTools分析软件进行比较分析。(2)以"结核"、"双向电泳"、"表面增强激光解析-电离飞行时间质谱"、"血清学"、"蛋白质组学"及相应英文为检索词在万方医学数据库、PUBMED中检索中、外文献进行文献复习。结果 WCX蛋白质芯片从血清中捕获到99种蛋白质/多肽,其中m/z为5 753.76的蛋白质在肺结核与肺外结核两组间差异有显著性,在肺外结核患者血清中表达上调,其平均质谱峰面积在肺结核患者和肺外结核患者中分别为15.42±4.86、28.29±14.08。结论结核病的发生可产生大致相同的血清蛋白质/多肽谱,在不同类型结核结核病患者血清中可以找到共同的血清学诊断标志物。Objective To study the expression of serum protein/polypeptides in patients with pulmonary tuberculosis so as to provide basic evidences for serological diagnostic.Methods of new type tuberculosis.Medthods（1） Serum protein/polypeptides mapping of 99 patients with pulmonary tuberculosis and 18 patients with extrapulmonary tuberculosis was performed using WCX protein chip and surface-enhanced laser desorption and ionization time of flight mass spectrometry（SELDI-TOF-MS）.The ClinProTools was introduced for comparative analysis.（2） keywords ＂tuberculosis＂,＂two-dimensional electrophoresis＂,＂SELDI-TOF-MS＂,＂serology＂,＂proteomics＂ and other related terms were used to search in Wanfang medical data base and PUBMED for Chinese and English literatures.Results Ninety nine proteins/polypeptides were captured by WCX protein chip,among which there was a significantly different expression of m/z（5 753.76） between pulmonary tuberculosis group and extrapulmonary tuberculosis group.The expression of m/z（5 753.76） in the serum of patients with extrapulmonary tuberculosis was up-regulated.The average peak areas in mass spectrum（MS） in the pulmonary tuberculosis group and the extrapulmonary tuberculosis group were 15.42±4.86 and 28.29±14.08,respectively.Conclusion Roughly the same serum protein /polypeptides mapping and the common serologic diagnostic markers can be found in the serum of patients with specific type of tuberculosis.
