摘  要:应用小鼠胚泡和外胎盘锥体外培养的方法,研究了纤粘连蛋白对小鼠胚泡发育及胚泡或外胎盘锥粘附和扩展的影响。结果显示,纤粘连蛋白对小鼠胚泡发育有一定的促进作用;对胚泡及外胎盘锥的粘附和胚泡初生滋养层细胞及外胎盘锥次生滋养层细胞扩展均有显著促进作用。纤粘连蛋白分子活性位点的合成肽段精-甘-天冬-丝氨酸可有效抑制纤粘连蛋白对胚泡或外胎盘锥发育、粘附和扩展的促进作用。结果表明,纤粘连蛋白在小鼠胚胎发育和着床过程中起着重要作用,这种作用是通过纤粘连蛋白分子活性位点对胚泡或外胎盘锥细胞上纤粘连蛋白受体的识别和结合实现的。Effects of fibronectin (FN) and its synthetic active peptide (Arg-Gly-Asp-Ser,RGDS) on mouse embryo development, blastocyst attachment and outgrowth on uterine epithelial cell monolayer or ectoplacental cone (EPC) on FN were studied by using in vitro culture models. The results showed that FN not only remarkably improved embryo development but also promoted attachment of blastocyst and EPC as well as outgrowth of primary and secondary trophoblast giant cells in vitro. On the other hand, these effects were inhibited by RGDS. These data suggest that FN plays a very important role in mouse embryo development and implantation and that this role is achieved by the recognition and binding between the FN active sites and FN receptors on the cell surface of embryo and ectoplacental cones.
