摘 要:目的:探讨肾综合征出血热(HFRS)毒株及病人血清分型方法。方法:快速荧光灶抑制试验(RFFIT)。结果:6株来自黑线姬鼠和病人的毒株,其免疫血清对Ⅰ型标准株76-118的中和效价较Ⅱ型标准株(UR)高4~256倍;而另6株来自褐家鼠和大白鼠的毒株,包括Gou3和K24的免疫血清,对Ⅱ型标准株UR的中和效价高于Ⅰ型76-118株8~128倍;被检的所有毒株和病人血清都能准确地分型。结论:RFFIT具有快速、简便、特异和准确灵敏的特点,不仅可检测中和抗体,且适用于HFRS病毒血清学分型。Serotypic classification of 12 HFRS viruses isolated from different sources and patient sera from different areas was studied using rapid fluorescent focus inhibition test (RFFIT).Sera of rabbits inoculated with different HFRS viruses were tested against reference type-ⅠHantaan virus and type-Ⅱ Seoul virus .The neutralizing titers of antisera of Apodemus isolates and patient blood against type-Ⅰ Hantaan virus were 4~256 fold higher than those to type-Ⅱ seoul virus,while the antisera of Rattus isolates-had titers to type-Ⅱ viruses 4~128 fold higher than those to type-Ⅰ viruses.So different isolates could be classified into serotype-Ⅰ and serotype-Ⅱ.In this study,we also found Gou 3 and K 24 ,which could be classified into serotype Ⅱ according to differences (≥4-fold) of the neutralizing titers between the sera reacted against reference type-Ⅰ and type-Ⅱ viruses, which is consistent with data previously published.Otherwise, serotypic classification of patient sera from different areas was investigated. The results showed that the RFFIT is an easy, fast、 specific and exact method and may be used in Serotypic classification of HFRSV.
